Variation in rhizobia strains isn’t a desirable fact based mainly on the possibility of unexpected results on legume inoculation. In this work, we studied the variability on phenotypic characteristics and genetic stability of rhizobia strains recommended for soybean inoculation. Variants with stable colony morphology were obtained from Bradyrhizobium japonicum strain SEMIA 5080 and from B. elkanii SEMIA 5019. Variants from SEMIA 587 obtained by another author were also used. The variants differed on colony characteristics, nodulation capacity, nitrogen fixation efficiency and competitive ability for nodule formation in two soybean varieties (Jacui 7 and IAS 5). Clonal characterization of Streptococcus mutans strains by multilocus enzyme electrophoresis BJM

Twenty-one Streptococcus mutans strains were clustered by Multilocus Enzyme Electrophoresis (MLEE). Six isoenzymes showed strong infra-specific discriminatory power (M1P, MPI, PLP, NSP, GOT, and LAP). MLEE is a robust technique that may be used to explore clonal diversity of S. mutans isolates in epidemiological surveys.

Total proteins and esterases from silk gland extracts of Bombyx mori silkworm were characterized and compared with electrophoretic profiles of prepared extracts with silkworm glands infected with nucleopolyhedrovirus (BmNPV). SDS-PAGE (7%) gels was used for the total proteins, and 10% native PAGE for esterases. In the silk glands extracts of healthy silkworms, it was observed seven protein zones with molecular weight varying between 10 kDa (P1) and 60 kDa (P7). In the infected silkworms, a new zone named P8 (90 kDa) was also detected. ABSTRACT Coffee is one of the main agricultural commodities in the country, and it is important to conservation of plant material for breeding programs. Cryopreservation is a promising alternative for preserving in the long-term the germplasm of species considered recalcitrant. However, studies should be performed to achieve maximum survival of seedlings after immersion in liquid nitrogen. This study aimed to investigate the desiccation tolerance of Rhamnidium elaeocarpum Reissek seeds through physiological and biochemical alterations. In the present work, isoenzyme electrophoresis was used to analyze the variability and phenetic relationships among seven American species of genus Prosopis belonging to three different sections: P. argentina (Monilicarpa), P. glandulosa, P. velutina, P. flexuosa, P. ruscifolia, P. kuntzei (Algarobia), and P. reptans (Strombocarpa). Identification of Leishmania (Leishmania) chagasi isolated from healthy skin of symptomatic and asymptomatic dogs seropositive for leishmaniasis in the Municipality of Rio de Janeiro, Brazil BJID

Euthanasia of seropositive dogs has been one of the principal measures adopted by the Program for the Control of Leishmaniasis in Brazil for many years. However, its efficacy is currently being questioned. We obtained intact skin samples from 20 Leishmania-reactive dogs from the municipality of Rio de Janeiro that had been referred for euthanasia. The promastigote forms of Leishmania were isolated in culture from 18 of these animals. Fourteen of these isolates were identified as Leishmania (Leishmania) chagasi by isoenzyme electrophoresis; seven of these were from asymptomatic dogs and seven were from symptomatic animals with visceral leishmaniasis. The internal morphology of embryos from immature and mature fruits of Heliconia bihai (L.) L. cv. Lobster Claw Two was examined. Embryos were inoculated into MS media (full MS and ½ MS) and GA3 (0, 2.5 and 5 mg L-1) with either sucrose or glucose. These plantlets were then replicated and transferred to MS medium (full MS or ½ MS) with 0 or 2.5 mg L-1 BAP and their multiplication was evaluated 30 and 45 days after inoculation. The genetic variability of the multiplied plants was estimated using isoenzyme analyses. Isoenzymatic polymorphisms in urban populations of Drosophila willistoni RChHN

Natural populations of Drosophila willistoni collected both in urban sites and in the wild, as control, were analysed with respect to their isoenzyme polymorphisms in six loci of enzyme systems, through horizontal electrophoresis. The first pool of populations was collected and analysed during the years 1987 and 1988, and the other during 1992 and 1993, in comparable seasons. The level of enzymatic polymorphism first detected was smaller in comparison to that observed four years later, suggesting an increase of variability along the time elapsed since the first sampling of this species, in the city of Porto Alegre. Molecular weight estimation of esterase isoenzymes in closely related Drosophila Species (Diptera: Drosophilidae) in non-denaturing polyacrylamide gel electrophoresis BABT

A method that allows the measure of molecular weight of two well-known and closely related esterases from Drosophila mojavensis and its sibling species, D. arizonae, is here described, using native polyacrylamide gel electrophoresis at several concentrations, applying Fergunson´s principles. These enzymes, namely EST-4 and EST-5, presented molecular weight values between 81 and 91 kDa. Components with phospholipase A2 activity were isolated by gel filtration and cationic exchange chromatography from the venom of Bothrops asper snakes from Chiriguaná, Colombia (9°22´N; 73°37´W). Five fractions were obtained by the gel filtration, and PLA2 activity was found in fraction 3 (F3). In the cationic exchange chromatography, F3 showed eight components with PLA2 activity. Six of these components appeared as one band in polyacrylamide gel electrophoresis (SDS-PAGE). Fractions II and VII exhibited an optimal activity at pH 9 and 52ºC. The enzyme chitinase from Moniliophthora perniciosa the causative agent of the witches' broom disease in Theobroma cacao, was partially purified with ammonium sulfate and filtration by Sephacryl S-200 using sodium phosphate as an extraction buffer. Response surface methodology (RSM) was used to determine the optimum pH and temperature conditions. Four different isoenzymes were obtained: ChitMp I, ChitMp II, ChitMp III and ChitMp IV. Structural and regulatory differences in amylase isoenzymes in germinating Brazilian barley cultivars Genet. Mol. Biol.

The amylase electrophoretic patterns of 10 Brazilian brewing-barley varieties with different malting grades and diastatic power were analyzed during the 7-day germination which occurs during the malting process. Intra and inter-variety genetic variability was observed at both the structural and regulatory level. In the first few days after germination all varieties showed a few active loci, all of them with low activity.
